图书简介
This book provides an account of the disastrous story of a whole field of research which went astray in different directions, in attempts to solve a problem by using the wrong tools aimed at the wrong target.
It all started from the misinterpretation of Schellman’s experiments which were carried out in the mid-1950s, and which were later encapsulated by the so-called "Hydrogen-bond inventory argument." This has led Kauzmann to propose the hydrophobic effect, while dismissing the more important hydrophilic effects, in proteins.
In addition, as a result of the misinterpretation of Anfinsen’s hypothesis, as well as a misunderstanding of the Second Law, people attempted a futile search for the structure of proteins in the global minimum of either the Energy, or the Gibbs Energy Landscape.
Introduction: The Protein Folding Problem: Some of the Myths Associated with the Protein Folding Theories; From Frank and Evans’ Iceberg-Formation Conjecture to the Explanation of the Hydrophobic Effect; From Schellman’s Experiments to Fersht’s Hydrogen Bond Inventory Argument; From Kauzmann’s Conjecture to the Myth that the Hydrophobic Effect is the Dominant Factor in Protein Stability; From Levinthal’s Question to Solving the Levinthal \"Paradox\"; From Anfinsen Hypothesis to the Frenetic Pursuit of the Global Minimum in the Gibbs Energy Landscape; Some Candidates which could Potentially Evolve into New Myths; Appendices: Definition of the Solvation Process and the Corresponding Thermodynamic Quantities; Definition of the Structure of Water and Changes in the Structure Induced by a Solute; Le Chatelier Principle Applied to the Two Structure Model for Water; The Stability Condition for a Chemical Reaction; Solvation Gibbs Energy of Hard Sphere (HS) and the Work of Cavity Formation; Formal Expression for the Thermodynamic Quantities of Solvation of a Non-Polar Solute; The Solvation Entropy and Solvation Energy and Structural Changes in the Solvent; The Solubility of a Simple Solute in Water and Structural Changes Induced in the Solvent; Ben-Naim’s \"Pitfalls: \" Don Quixote’s Windmill; Examples of Gibbs Energy Landscapes and Gibbs Energy Functionals; The Single Minimum of the Gibbs Energy Functional; Non-Additivity of the Gibbs Energy Function; Non-Additivity in Binding Systems; Frustration in Three Dipoles on a Lattice, an Informational Theoretical Approach;
